Investigation of cytotoxic effect on human cervical cancer cells (HeLa) of chlorogenic acid

Abstract (EN)
In this study, chlorogenic acid (KA) different concentrations (100 µg/mL, 150 µg/mL, 300 µg/mL ve 500 µg/mL) alone and Tekamen (40 µg/mL) with different times (24, 48 and 72 hours) human cervical cancer (HeLa) cell line and human peripheral lymphocytes vitro cytotoxic effects MTT and WST-8 was determined with their viability tests. In this work HeLa that contain just medıum has been as a negative control and KA solution used as a control solvent containing %0.1 of DMSO and Tekamen been used for positive control.According to the result obtained in both experiments the reactins of chlorogenic acid in HeLa cell did not shows important cytotoxic effects. KA+Tekamen reaction has more cytotoxic effect in HeLa cell than its effects when used alone. Comparing the cytotoxic effects of KA+Tekamen in negative, positive and solvent control the statistical differencies were found significant. (p<0.05). KA alone does not have important cytotoxic effects in peripheral lymphocytes treatment, KA+Tekamen reactions shows more cytotoxic effects than when used alone. Comparing the reaction KA+Tekamen treatment with negative control, positive control and solvent control the differencies has been statistically significant (p<0.05). The result of KA either or KA+Tekamen mixture reaction, shows that the % cytotoxic in HeLa cells including datas has been higher than the datas obtained in viability test of MTT and the result obtained in WST-8 viability test. In the same way KA alone and mixture of KA+Tekamen reactions the result of viability tests in peripheral lymphocytes MTT and WST-8 appliyed to different periods has given different results.
