5-aminolenulinic acid mediated photodynamic diagnosis in colon and gastric cancer cell lines

Today, colon and gastric cancers are common cancers in all types of cancer. The methods used for early detection of these cancers do not provide enough information as necessary. For this purpose, photodynamic diagnosis has started to be used recently. In this study; it was aimed to determine the conditions required for the most effective photodynamic recognition of the fluorescence produced by 5-ALA stimulated PpIX in colon cancer (HCT116) and gastric cancer (AGS and HCT116) in vitro. In this thesis, HCT116, AGS and MKN28 cell lines which were administered different 5-ALA (0, 100, 200, 300 300 μg / ml) concentrations and incubations time (0, 1, 2, 3, 5 hours), were performed on fluorescence images obtained by cell viability, fluorescence intensity analysis, cell migration and mobility analysis by imaging processing, early cell apoptosis analysis with caspase3/7. When the cells were stimulated by 200 μg / ml 5-ALA on 3 hours, the cell viability of AGS cell lines decreased more than the cell viability of MKN28 and HCT116 cell lines. Different fluorescence intensities of cancer cells were determined at various incubation times and 5-ALA concentrations of MKN28, HCT116 and AGS cell lines. The most pronounced fluorescence intensity was observed on the MKN 28 cell line. It was determined that the highest cell movement was in the MKN-28 cell line and the lowest cell movement was in the HCT116 cell line. Early detection of apoptosis in the HCT116 cell line stimulated with 300 μg / ml 5-ALA was more pronounced when compared to other cell lines. As a result, fluorescence observed after 3 hours in cells stimulated with 200 μg / ml 5-ALA was found to be the most suitable diagnosis for photodynamic diagnosis of gastric and colon cancers. Photodynamic diagnosis, which will show efficacy and sensitivity in future human studies, can be applied as a promising method in the early diagnosis of gastric and colon cancers clinically.
