The investigation of PON1 and PON2 polymorphisms planning in CABG patients

Abstract (EN)

PURPOSE: Coronary artery disease (CAD) is one of most important health problems in developed countries. Hypertension and type 2 diabetes mellitus (T2DM) cause endothelial dysfunction probably through increased oxidant stress. Thus hypertension and type 2 diabetes mellitus (T2DM) play role in the etiopathogenesis of CAD. Paraoxonase (PON) is a high-density lipoprotein-bound anti-oxidant enzyme that inhibits atherosclerosis and endothelial dysfunction. In this study, We scheduled to investigate the relationship between PON1, PON2 and PON activity in patients with T2DM or HT.METHODS: Ninety-two patients were enrolled in the study. They were divided into three groups. Thus, patients were separated as group T2DM (n=31): group HT (n=30) and group control (n=31) were composed of individuals healty patient. PON1 Leu 55 Met, Gln 192 Arg; PON2 Ser 311 Cys polymorphisms and serum PON activity were evaluated by PCR-RFLP techniques and measuring spectrofotometric paraoxon hydrolysis.respectively. The association between PON1, PON 2 ,PON activity and lipid profiles were evaluated in the CAD.RESULTS: PON1 and PON2 genotypes of all three groups the allele frequencies and genotype distribution were not significant differences in terms of looking at the PON1 L55M, PON1 Q192R and PON2 C311S genotype homozygous codon among three groups (P>0.05). When compared control group PON1 activity was found higher in the T2DM patients. This difference was not significant. However, when compared control group with the HT group was higher. PON activity difference was significant (p<0.05). Serum PON activity was a significant correlation with other biochemical parameters (P<0.05).DISCUSSION and CONCLUSION: These findings suggest that serum PON activity is reduced in the T2DM patients. However, serum PON activity is higher in the HT patients. In conclusion detection of changes in PON 1, PON 2 polymorphisms and activity of serum PON1 enzyme a complex and multigenic disease at an early stage will provide the opportunity precaution in the diagnosis and treatment of CAD at-risk individuals.
