Association of IL-10, IL-17A, IL1-7F, and TGFß1 gene polymorphisms with chronic lymphocytic leukemia

Abstract (EN)

Aim: In this study we aimed to determine the distributions of genotypes and allele frequencies of IL-10 (-592A/C), IL-17A (-197A/G), IL-17F (7383A/G) and TGFß1 (+915G/C) polymorphisms and revealed association between chronic lymphocytic leukemia and these polymorphisms.Materials and Methods: We isolated DNA from peripheral blood and genotyped the SNPs of IL-10 (-592C/A), IL-17A (-197A/G), IL-17F (7383A/G) and TGFß1 (+915G/C) by using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis in 50 CLL patients and 50 healthy controls.Results: The distributions of genotypes and alleles in CLL patients and healthy controls were compared. There wasn?t statistically significant relationship between susceptibility to CLL and IL-10 (-592A/C), IL-17A (-197A/G) , IL-17F (7383A/G) and TGFß1 (+915G/C) polymorphisms. Also we investigated the association between these polymorphisms and clinical characteristics and laboratory findings of patients, such as age at diagnosis, gender, lymphocyte count, Rai stage, FISH findings. Patients with TGFß1(+915GC) genotype have a higher frequencies of trisomy 12. And, female patients have a higher incidence of IL-17A (-197AG) genotype than male patients.Conclusions: In conclusion, our data don?t support the role of IL-10 (-592A/C), IL-17A (-197A/G), IL-17F (7383A/G) and TGF?1 (+915G/C) polymorphisms for CLL susceptibility. However, TGFß1 +915GC is found associated with trisomy 12 which is predisposed to longer overall survive compared to the normal karyotype. And females with IL-17A -197AG genotype should be more susceptible to the CLL compared to the males with IL-17A -197AG genotype.
