Investigation of SETD2 gene expression in patients with laryngeal cancer

Abstract (EN)
Laryngeal cancer is a rare type of tumor in which malignant cells grow in the larynx. Cigarette and alcohol consumption are the main risk factors for laryngeal cancer. Apart from the vital functions of the person it is also important for the social life of the person due to the location of the larynx in the body. For this reason, it is aimed to protect the laryngeal resection as well as the duration of the patient's survival. The difficulty of diagnosis and the determination of the disease mostly in advanced stages have accelerated the studies that can provide early diagnosis of laryngeal cancer. Despite all developments, recurrence and treatment of the disease resistance still make treatment difficult. Laryngeal cancer is still in the group of cancer patients who are difficult to treat. For these reasons, biomarkers and more effective methods of treatment that can be used in the early stages of aging are needed nowadays. There are no approved tests to detect malignant tissues and confirmed high specificity to detect early stage detection for.laryngeal cancer. It is very important that oncology studies can compare expression levels between tumor cells and normal cells. The SETD2 gene is located on the cytogenetic band p21.31 of chromosome 3, a region frequently targeted by loss of copy number in various tumors. SETD2 plays a role in DNA repair, chromosome segregation and RNA addition. Therefore, we aimed to investigate the expression levels of the SETD2 gene which can be used as a marker in larynx cancer, diagnosis and follow-up in our thesis study. As a result of the study, patient tumor samples, conjugated healthy normal tissues and serum samples collected before surgery were compared and evaluated in terms of SETD2 gene expression. In 32% of patients (16/50) there was an increase in the level of expression compared to the normal tissue in the tumor,26% (13/50) of patients showed a decrease in tumor expression level. When the expression status and the clinicopathological data of the patients were compared, no significant difference was found between smoking and alcohol consumption, recurrence, anatomic involvement zones and metastasis (p> 0.05). Only a significant correlation was observed between expression increase in the SETD2 gene and lymph node involvement (p = 0.048).
