Abstract (EN)

Carbon dots have distinctive properties such as good biocompatibility, non-toxicity, high mechanical/thermal properties and relative ease of functionalization. Their easy, one-step synthesis from biological materials is attracting increasing attention compared to toxic chemicals and energy-intensive traditional approaches. Our aim is to synthesize N-doped carbon dots with low toxicity and high antioxidant and antimicrobial effects from Laurocerasus officinalis leaf extract by hydrothermal method for use in cell imaging. Again, it is aimed to evaluate the cytotoxicity of these synthesized carbon dots in lung cancer cells (A549) and to visualize them with a confocal microscope. The cytotoxicity of carbon dots in A549 cells was measured by WST-8 analysis. Antioxidant activity was determined by DPPH and antimicrobial activity was determined by disk diffusion method. As a result of the analysis, it was shown that the synthesis carbon dots showed cytotoxicity (anticarcinogenic activity) in A549 cells only at high concentrations (250-500 ug/mL), while they increased cell viability at low concentrations. Again, it showed antioxidant activity at high concentrations (200 µg/mL and above). Carbon dots have shown strong antibacterial and antifungal activity. Carbon dots have shown strong antibacterial and antifungal activity. Additionally, carbon dots have been proven to enter the cell nucleus by DAPI staining. The fact that the carbon dots we synthesized as a result of these studies have strong anticancer, antimicrobial and antioxidant properties and reach the cell nucleus has provided them with the potential to be used especially in imaging cancer cells and in cancer treatment.
