Abstract (EN)

In recent years, there has been a significant global increase in the use of synthetic cannabinoids (SCs). These compounds, marketed under various brand names such as "Bonzai," "Spice," "K2," "Spice Gold," "Spice Diamond," "Chill Out," and "Chill X," are among the most commonly encountered psychoactive substances posing a threat to public health. Their widespread use can be attributed to their low cost, rapid and potent psychoactive effects, and the fact that they remained undetectable by conventional analytical methods for an extended period. In this thesis study, a novel and reliable liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the determination of two frequently encountered SCs, MDMB-4en-PINACA and ADB-BUTINACA, in blood and urine samples. To eliminate matrix interferences during sample preparation, solid-phase extraction (SPE) was employed. The developed method demonstrated excellent linearity within a concentration range of 1.0–50.0 ng/mL, with a high correlation coefficient (r² = 0.998). High recovery (RE) rates and the successful fulfillment of all necessary validation parameters for forensic toxicological analysis confirmed both the analytical reliability of the method and its applicability to real forensic samples. In this study, MDMB 4- en Pinaca and ADB Butinaca, which have been increasingly used in recent years, were analyzed in blood and urine samples by solid phase extraction method using LC-MSMS. In the measurements, the linear range for all analytes was determined as 1.0-50.0 ng/mL with a correlation coefficient higher than 0.992. The detection limit in blood and urine samples was found to be 0.3-0.5 ng/mL, and intra-day and inter-day reproducibility was found to be below 10%. In our study, where high recovery values were obtained, quantitative determination was performed in blood and urine samples with validation parameters suitable for toxicological analyzes.
