Abstract (EN)

In this study, prokaryotic diversity of Meke Lake and Acı Lake (Konya) was investigated.Samples were taken from the Meke Lake and Acı Lake (Konya) on August 2010 and April 2011. Isolation was performed on Medium A, B, C, D, E and %12, %18, %23 MGM. 16S PCR was carried out for pure to colonies on media and amplified ribosomal DNA restriction analysis (ARDRA) was applied the 16S PCR products. The nucleotid sequences of the purificated PCR products were sequenced and 4 different strains in 46 isolates for Acı Lake and 13 different strains in 220 isolates for Meke Crater Lake were identified. 16S rRNA gene sequences were compared with reference sequences at NCBI using BLAST. According to the BLAST results, Halomonas, Marinobacter, Idiomarina, Thalassospira in Bacteria domain for Acı Lake, Salinibacter, Salinivibrio, Halomonas, Idiomarina and Marinobacter in Bacteria domain and Halorubrum in Archaea domain for Meke Lake were identified.Denaturing gradient gel electrophoresis (DGGE) was performed on samples for purposes that were studying population dynamics, monitoring enrichment cultures and determining rRNA operon copy number. Enrichment cultures of DGGE showed that there are Halorubrum sp. and Haloterrigena limicola in Meke Crater Lake as culturable species. Dominant species were identified on August and April samples of Meke Crater Lake by DGGE. Sequence analysis showed existence of uncultured Salinibacter sp. was dominant in Bacteria domain and uncultured archaeon was dominant in Archaea domain for August sample. Similarly, uncultured archaeon was dominant in Archaea domain and uncultured bacterium was dominant in Bacteria for April samples. 11 bacterial isolates were used for determination of rRNA operon copy number on DGGE and results showed that some of bacterial species have 16S operon copy number more than one.Fluoresceine in situ hybridization (FISH) was used both Acı Lake samples and Meke Lake samples. The community was dominated by Archaea on the Meke samples. Acı Lake was showed only bacterial fluorescent hybridization.Prokaryotic diversity of Acı Lake and Meke Lake was first time characterized and existence of some strains were showed first times in Turkey. Keywords: Meke Crater Lake, Acı Lake, ARDRA, DGGE, FISH.
