Investigation of MDR 1 gene C3435T, T1236C, G2677T, A2956G polymorphisms in breast cancer cases

Cancer is a disease that causes changes and uncontrollable reproduction of cells in living bodies and it is on the top of the disease list that causes to death. Breast cancer is the most common cancer type for woman. Although some important steps were taken for breast cancer medication, chemotherapy which is the most important weapon for breast cancer treatment does not give successful results in some cases. It is considered that MDR1 gene has an important role on susceptibility to this disease and drug resistance developed against medicines used in cancer and some other diseases. In addition to that, polymorphisms of this gene shows differences among the ethnical groups and its effects changes among these groups. In this study, it is aimed to compare C3435T, T1236C, G2677T and A2956G polymorphism of MDR1 gene among a study group which consist of 35 patient with breast cancer and 20 healthy people and determine whether there is a relation between breast cancer and these polymorphisms. In addition, the incidence of these polymorphisms was determined for Turkish population. During the studies, DNA isolation was performed for collected blood samples and genotypes of study group for these polymorphisms were determined by using PCR (Polymerase Chain Reaction) - RFLP (Restriction Fragment Length Polymorphism) method. SPSS program was utilized for statistical evaluation of experimental data. Results showed that there is no meaningful difference between patient and control group in terms of T1236C, G2677T and A2956G polymorphisms. But TT genotype incidence is signifcatly high for C3435T polymorphism in patient group and it can be responsible for susceptibility to breast cancer.
