Expression of nephronectin in metastatic and non metastatic tumors

Abstract (EN)

Breast cancer, which can cause of death if its not treated, is commonly occurs in women . One of the reason of breast cancer as a dangerous illness high quality of the its metastatic ability. Metastasis means that; a tumor split from an organ and create a new tumor one another organ. For existing metastasis firstly tumor grow up in settled tissue and then migrate to a new organ via blood vessels. During the tumor metastasis, tumor create a new environment with disturb to normal tissue structure a new tissue. So during the reprogramming to extracellular matrix proteins also play a role. Nephronectin, an extracellular matrix proteins and as a play a role epitelial mesenchymal transition. There is no working about the effect of nephronectins which can take a role in tumor grows on metastatic tumors. So, in this study we aimed to determine the nephronectin expressions by using metastatic and non-metastatic cell lines in primary tumors and metastasis. In our study, 4TLM and 4THM cell lines, known as metastatic, and 67NR cell lines, known as non-metastatic, is ortotopicaly injected to mice. Primary tumors, lung and liver tissues, taken in 12th and 25th day, is prepared for light microscopy and western blot investigations. Immunohistochemistry methods are implied for nephronectin and Ki67 proteins in tissue sections; and localization of this proteins are determined. The protein expression levels of nephronectin and Ki67 were also investigated by western blot. According to our results, decreased of nephronectin expressions were observed in primary tumors, lung and liver in 4TLM and 4THM injected mice. Moreover there is a notablu decrease of nephronectin expressions 25thday ,high level of metastasis, than 12th day, not yet metastasis. Ki67 as a proliferation marker is highly localizated in metastatic cell lines tissues. It is also seemed not highly, in non-metastatic cell line tissues. Ki67 expression levels is clearly observed in tissue taken from 25th day than the tissue taken of 12th day. In our study we showed for the first time in literature that the expression levels of nephronectin demonstrated in primary tumors, lung and liver tissues which is obtained from metastatic cell lines. Result of our study shows that the possibility of that cancer cells can disturb the localization of nephronectin proteins in extracellular matrix for invasion to tissue and reprogramming extracellular matrix to metastasis. Accordingly nephronectin is considered as a protein which play a role in during metastasis process.
