Abstract (EN)
Methimazole is one of the first-choice antithyroid agent used especially in the treatment of hyperthyroidism and Graves disease. In addition to the known adverse effects of methimazole, such as nausea, vomiting, and urticaria, pancreatitis case reports related to the this active substance have been reported through literature studies. Although data on the risk of drug-induced acute pancreatitis are mostly based on case reports and information obtained as a result of case control, animal and in silico studies; in vitro studies to elucidate the mechanisms of formation of this risk are scarce. It is foreseen that in vitro studies to clarify the mechanisms of pancreatitis induced by drugs will be very useful in terms of eliminating the deficiency in this field, and our thesis study aimed to define the toxic effect mechanisms underlying metimazole induced pancreatitis. In this study, IC30 value was determined and it is thought that the formation of reactive oxygen species may increase the risk of methimazole-induced pancreatitis. The results obtained are intended to contribute to the literature and provide a basis for the planning of further new researches on the subject.
