Abstract (EN)
Objective: Meyerozyma guilliermondii is a species of yeast that causes serious infections such as bloodstream infection (candidaemia) as an opportunistic human pathogen. In this study, it was aimed to determine the appropriate restriction enzyme for Pulse Field Gel Electrophoresis- Restriction Enzyme Analysis of Genomic DNA (PFGE-REAG) method which can be used in molecular epidemiological analysis of M. guilliermondii strains. Materials and Methods: In this study, epidemiologically related 12 M. guilliermondii strains of 9 patients isolated in the Medical Microbiology Laboratory of the Medical School Hospital of the Karadeniz Technical University were included. PFGE-REAG method was studied by using CHEF DR III (BioRad, USA) device system with SfiI, NotI and PacI restriction endonuclease enzymes. Results: As a result of restriction reaction with SfiI, bands that were scattered across the range of molecular size markers and were more clearly distinguishable were observed. SfiI and NotI band profiles were compared with the karyotyping band profiles; It was concluded that the REAG protocol was more distinctive than the karyotyping. Conclusion: REAG-SfiI can be used to determine the clonal relationship of epidemiologically related M. guilliermondii strains. This study contributed to the determination of the REAG method which will help to evaluate the clonal relatedness of epidemiologically related M. guilliermondii clinical isolates.
