Abstract (EN)

Acute Myeloid Leukemia (AML) is characterized by differentiation and overgrowth of blast cells. It was aimed to investigate the role of miR-7 5p, which is known to act as a tumor suppressor, through AML and its relationship with target genes. For this purpose, miR-7-5p mimic was transfected to HL60 and NB4 AML cells by Lipofectamine-mediated method. qRT-PCR was performed for transfection validation. The effect of miR-7-5p on proliferation was investigated by WST-8 method, and its effect on apoptosis was investigated by Caspase 3 assay. The relative gene expressions of BCL2, SKP2, OGT, KLF4, EGFR genes which were determined as possible target genes of miR-7-5p using the miRDB, miRTarBase, Targetscan and miRWalk databases and VIM gene known as the EMT marker were investigated by qRT-PCR technique. In the miR-7-5p transfected group, compared to the control group, cell proliferation was found to be significantly decreased and apoptosis increased. It was determined that the expression levels of SKP2, KLF4 and OGT genes were statistically decreased in the miR-7-5p transfected group. However, it was detected that BCL2, EGFR and VIM genes expression levels did not change statistically. In addition, it was thought that miR-7-5p might affect the cancer process in AML by targeting SKP2, KLF4 and OGT genes. It is very important to detect and confirm the target genes of miR-7-5p, which has the potential to be a new marker in the early diagnosis and treatment of AML. Therefore, our data obtained at the mRNA level should be confirmed by further studies.
