Abstract (EN)

Alzheimer's disease is a progressive, fatal form of dementia of unknown cause, usually seen in the elderly population, characterized by behavioral symptoms and loss of cognitive and physical function. Alzheimer's disease is increasing every year. Since the etiology of the disease is not fully known, the treatment is usually continued to alleviate the symptoms and to counteract the progression to the advanced stage. Various AChE and MAO inhibitors are used in the symptomatic treatment of AD. In this study; 10 thiazole-hydrazone derivative compounds were synthesized. The obtained compounds were analyzed by 1H-NMR, Mass spectroscopic methods. Inhibitory effects of synthesized compounds on cholinesterase and monoaminoxidase (MAO) enzymes were investigated. No significant activity was detected against the butyrylcholinesterase (BChE) enzyme in any of the synthesized compounds. But 3b, 3e, 3i and 3j coded compounds; They showed remarkable effects on AChE, MAO-A and MAO-B enzymes. Within the scope of the thesis, the structure of AChE, MAO-A and MAO-B enzyme active sites was clarified with molecular modeling studies, docking studies were performed on active compounds, and enzyme active site and binding points were determined. It was revealed that the strongest interaction with AChE, MAO-A and MAO-B enzyme active sites was observed with the 3b coded compound.
