Abstract (EN)

The neo-natal FcRn receptor is a protein that in humans is encoded by FcGRT gene and has 46 kDa molecular weight. It is also known as IgG-Fc receptor and is similar to major histocompatibility complex (MHC) class I. In early studies, FcRn is the only receptor which plays an active role in IgG circulation realized between mother and fetüs. Moreover, FcRn receptor binds to Fc domain of IgG with high affinity to FcRn at acidic pH (<6.5) but not at physiological pH (7.4). For this reason, separation and purification of FcRn, the development of drug delivery, separation and diagnostic platforms which are structure immunoglobulin and alternative technological researches.In this study, P(IgG-co-HEMA) cryogel column has been prepared for the separation and purification of neonatal FcRn receptor from the human placenta. In this direction, the sections that have, extracted from epitelium cells of placenta, especially from syncytiotrophoblasts, have homogenized by Triton X 100. Then, these paration of FcRn receptor has carried out by Fast Protein Liquid Chromatography (FPLC) using P(IgG-co-HEMA) cryogel column starting point that the interaction of IgG-FcRn receptor realizes at acidic pH but not at physiological pH.
