Combined use of quercetin and ficetin flavonoids with pregabalin and gabapentin in the treatment of neuropathic pain

Abstract (EN)

Quercetin and fisetin are flavonoids that are known to exert various pharmacological effects, including antinociception, and their effect on neuropathic pain has not been studied in detail before. In this thesis, it was aimed to investigate the effects of quercetin and fisetin alone and in combination with gabapentin and pregabalin, which are used in neuropathic pain treatment, on neuropathic pain. The anti-allodynic effect of quercetin and fisetin at various doses (5, 10, 20 mg/kg), both alone and in combination with pregabalin and gabapentin, was evaluated by developing a neuropathic pain model induced by chronic constrictive nerve injury in rats. Both flavonoids showed a significant effect compared to the control group at doses of 10 and 20 mg/kg. The efficacy of these flavonoids was investigated in combination with gabapentin (50 mg/kg) and pregabalin (15 mg/kg) by selecting an effective dose of 10 mg/kg and doses of 5 mg/kg without significant antiallodynic effects. According to the data obtained; In our groups combined with gabapentin and pregabalin, it was determined that they showed significant antiallodynic effects compared to 50 mg/kg gabapentin and 15 mg/kg pregabalin. As a result; In our combination studies, it has been observed that the efficacy of gabapentin and pregabalin, which are frequently used in the treatment of neuropathic pain, when used together with flavonides at lower doses, increases the effectiveness and prolongs the duration of action.
