Abstract (EN)

The enzyme that work for cleavage of hydrogen peroxide which is one of the reactif oxygen specieses to water and molecular oxygen is catalase and it is similar to oxygen bridged dinuclear manganese complexes regarding to its structure. For this reason, the synthesis of structures as catalase enzyme has an increasing interest. In this systems, it is known that the structures of the complex affect activity of manganese. In this thesis, dinuclear manganese complexes with mixed ligands ([Mn2(µ-2-NH2C6H4COO)2(phen)4]•2(ClO4)(CH3OH) (1), [Mn2(µ-2-NH2C6H4COO)2 (bipy)4]•2(ClO4) (2) and [Mn2(µ-4-FC6H4COO)4(H2O)(bipy)2] (3)) which are similar to catalase enzyme were synthesized and characterized. The reaction conditions were optimized in variable mediums like solvent, temperature and base for activity of cleavage of hydrogen peroxide (catalase activity) of this structures. Reaction rate equation for disproportination of peroxide was determined as ʋ= 47,8[H2O2]0,7[catalyst]1,9 by kinetic studies. Interaction between metal complexes and DNA has is a multidicipliner research topic in chemistry, molecular biology and medicine. Interest in this field have increased fastly since utilizing of cis-platin [Pt(NH3)2Cl2] for inhibition of cell division and chemotherapy, development in metal mediated drugs. Utilizing of traditional platin mediated molecules is limited to inhibition by little cellular molecules and throwed out of cell. These limitations require to develop more affective, less side effects and special targeted DNA-binding drugs. In this study, the DNA binding activity of synthesized complexes were investigated in different DNA and complex concentrations. Electronic absorption titration and floresance quenching methods were used to investigate the activity of metal-DNA binding. Conclusion of measurements, DNA binding affinity of complexes was found as 3>1>2. Key Words: DNA, catalase, complex, chemotherapic, manganese, toxic
