The protective effects of resveratrol in the treatment of acute poisoning with organophosphate compounds

Abstract (EN)
In this thesis, the antioxidant and protective effects of resveratrol (RES), pralidoxime (PAM) alone and together in different doses against oxidative damage induced by acute fenthion toxicity were investigated by biochemical, immunohistochemical and histopathological methods in rats. After fenthion-induced acute toxicity, malondialdehyde (MDA), reduced-glutathione (GSH), ascorbic acid, retinol, β-carotene levels in blood samples, erytrocyte superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) enzyme activities and liver, heart, kidney, brain, lung tissue MDA, GSH levels were investigated by biochemical methods. Furthermore immunohistochemical and histopathological examinations of liver tissues were studied. It was observed that LPO marker MDA was increased in all toxication groups. In all treatment groups RES and PAM decreased the MDA levels and reduced fenthion-induced LPO. RES and PAM prevented the loss of retinol and β-carotene levels. There was no significant difference in the SOD, GSH-Px and CAT activities in treatment groups. It was observed that fenthion increased the LPO in liver, brain, kidney and heart tissues. However RES decreased the LPO in liver, brain and heart tissues. It was obtained from the histopathological examination of the liver that the lesions occurred in the intoxication group were decreased in treatment groups. Immunohistochemical analysis showed that poisoning didn't reach up to apopitosis level in all groups. However in the immunohistochemical stainings, RES prevented apoptosis. PAM was at least as effective as RES. As a result, RES prevented the LPO and oxidative damage induced by fenthion and showed protective effects in the liver tissues against the damage caused by OF poisoning.
