The effect of organophosphorus compound glyphosate on oxidative stress parameters in in vitro cord blood erythrocyte cells

Abstract (EN)

Aim: In order to further define the role of glyphosate, a broad spectrum organophosphorus herbicide used against weeds, on oxidative stress of the cell and to better understand the effect of its exposure, the effect of in vitro on cord blood erythrocyte cells was investigated. In this context, it is aimed to determine total oxidant capacity (TOC), total antioxidant capacity (TAC), lipid peroxidation (LP) and osmotic fragility (OF) in umbilical cord blood erythrocytes treated with glyphosate at various doses. Material and Method: Fifteen healthy, non-smoking, cesarean women in the third trimester of pregnancy were included in the study, informed about the study and signed a consent form. After cesarean delivery, cord blood samples of pregnant women participating in the study were collected from the part of the cut umbilical cord on the placenta side, and quickly collected into tubes containing heparin using a sterile injector. Erythrocyte cell packs were prepared from the collected cord blood. Glyphosate in different concentrations (10, 100 and 500 μM) was applied to the erythrocyte suspensions and incubated for 3 hours at 37oC in glucose buffer. TOC, TAC, LP and OF analyzes of the prepared samples were made using spectrophotometric methods. Results: A statistically significant increase was found in the erythrocyte malondialdehyde and TOC levels in the sample groups after exposure to the cord blood erythrocyte cells at concentrations of 10, 100 and 500 μM glyphosate (P<0.05). However, cord blood erythrocyte TAC levels and OF values were not significantly different from the control group in the sample groups treated with 10, 100 and 500 μM glyphosate concentrations (P˃0.05). Conclusion: In this study, the effect of glyphosate, an organophosphorus herbicide, on oxidative stress parameters in in vitro cord blood erythrocyte cells was investigated. It was observed that glyphosate, an organophosphorus compound applied in different concentrations, increased LP and TOC levels, which are oxidative stress markers in xiv cord blood erythrocyte cells, but did not affect erythrocyte OF values and TAC levels in in vitro cord blood.
