Abstract (EN)

Pain is a type of symptom that is experienced frequently in life, occurs for different reasons and reduces the quality of life. Various treatment methods are available, but research continues intensively to find new pharmacotherapy methods that are natural in origin and have fewer side effects. In this thesis, it is aimed to report a study evaluating the effect profile of p-coumaric acid's known anti-inflammatory and antioxidant effects and its possible analgesic effects. In this direction, experiments were carried out with the whole cell configuration of the patch clamp method on primary dorsal root ganglion cells and the possible effects of p-coumaric acid on K+ currents were evaluated. Inhibition with 100 μM p-coumaric acid was observed in the depolarizing voltage range at which neurons were stimulated and in the delayed rectifier K+ current. These findings indicate regulatory effects on neuron excitability. This situation, observed on DRGs, one of the cornerstones of pain transmission, highlights the possible analgesic effect. The result of this study reveals the need to evaluate p-coumaric acid and its derivatives in pain studies. Keywords: Pain, Dorsal Root Ganglion, Patch Clamp technique, p-Coumaric acid.
