In vitro investigation of the effects of current paediatric restorative materials on human dental pulp stem cells

Abstract (EN)

This study aimed to evaluate the cytotoxic effects of the restorative materials currently used in pediatric dentistry on human dental pulp stem cells. The human dental pulp stem cells used in the study were obtained from embedded third molars, which were surgically extracted from individuals between the ages of 19-25. After the cells were isolated and confluent, passaging was started and the cells in the third passage reflecting the best cytogenetic properties of the cells were used in the study. The release liquid of GC Fuji IX GP, GC EQUIA Forte, APA Glass Carbomer and APA Esthetic Fill were exposed to the dental pulp stem cells. At the end of the 1st, 3rd and 7th days, cells were stained with Anneksin V and 7-Aminoactinomycin D (7-AAD), and live cells, necrotic cells, early and late apoptosis cells were analyzed by flow cytometry. Datas were statistically evaluated with IBM SPSS V23. When the data were evaluated, APA Glass Carbomer is the material with the highest live cell value in terms of cell viability and a statistically significant difference was found between other restorative materials (p<0.001). In terms of early apoptosis findings, there was no significant difference between the groups (p<0.05). Although there was no statistically significant difference between the restorative material groups in terms of late apoptosis findings (p<0.05), there was a statistically significant difference between the 3rd and 7th days for the GC EQUIA Forte material (p<0.001). In terms of necrotic cell findings, APA Glass Carbomer material has the lowest necrotic cell value and there is a statistically significant difference between other restorative material groups (p<0.001). As a result of the findings obtained from the study, APA Glass Carbomer material was determined to be a more biocompatible material at the end of the 1st, 3rd and 7th days compared to other restorative material groups.
