Preperation of novel polymers functionalizable with Ph-sensitive clevable covalent bonds

Abstract (EN)
In this study, it was aimed to prepare polymers containing functional groups on their side branches in such a way as to form silyl ether bonds with target molecules containing an alcohol group. For this purpose, new type of monomers containing a silyl ether chloride group were synthesized and these monomers were carried out by RAFT (Reversible addition- fragmentation Chain Transfer) polymerizations with hydrophilic monomers. The resulting functional polymers were connected to model molecules containing alcohol groups with silyl ether groups that can be hydrolyzed in an acidic environment. The monomers used were created in three different structures obtained by changing the alkyl groups on the silicon atom. Since the silyl ether group has a structure that is stable in basic environment but can be hydrolysed in acidic environment, small alkyl groups on the silicon atom allow silyl ethers to be hydrolysed more easily in acidic environment, while sterically more voluminous substitution groups allow silyl ether hydrolysis to be slower. Taking advantage of this difference, it was aimed that polymers functionalized with molecules containing an alcohol group release bound molecules by breaking the silyl ether group under acidic conditions. By preparing polymers containing reactive monomers in different proportions, it has been possible to bind target molecules to polymeric platforms at the desired level. Model molecules containing alcohol groups of different structure were attached to these new type of reactive polymers prepared. The optimum conditions for the synthesis and functionalization of synthetically reactive monomers and polymers have been investigated. The main purpose of the study is to prepare reactive polymers that can be functionalized by making a silyl ether bond that can be broken under acidic conditions with molecules containing an alcohol group, which is the main goal of the study. For this purpose, it was aimed to separate the bound molecules from the polymer as a result of hydrolysis of the silyl ether groups under appropriate conditions. Again, it was aimed to adjust the hydrolysis rates by changing the alkyl groups in the silyl chloride structure to be used as a reactive monomer. In addition to the subject of the study, a potential application area of this kind of functional polymer design could be the preparation of polymer-drug conjugates capable of controlled drug release.
