Abstract (EN)
Cancer ranks second after cardiovascular diseases in terms of death rates. Breast cancer is the most frequently diagnosed type of cancer and causes the most deaths among women. Polydatin (PD) is extracted from the Polygonum cuspidatum plant. PD has been shown to inhibit cell proliferation and induce apoptosis in tumors of various types of cancer. Long non-coding RNAs (LncRNA) mediate the formation and progression of cancer by epigenetically changing the expression of genes related to proliferation and apoptosis. In our thesis study, % viability and IC50 doses at 24 and 48 hours were determined in MCF-7 breast cancer cells applied with different doses of PD. In the LncRNA expression screening performed via microarray in MCF-7 cells applied with IC50 dose for 24 hours, the most dramatic change was found to be in LncRNA SLC9A3-AS1. The effect of LncRNA SLC9A3-AS1 on proliferation and apoptosis genes was examined by RT-PCR after silencing the LncRNA SLC9A3-AS1 gene. As a result of RT-PCR analysis, it was determined that the expressions of proliferation-related (JAK2, NRAS, TP73, RASSF1, BRCA1 and HGF) and apoptosis-related (TRAF2, AIFM1, APAF1, TP53, HRK, NFKB1 and CASP2) genes showed a statistically significant change in SLC9A3-AS1-siRNA transfected MCF-7 cell lines (p< 0.05).It was determined that the number of apoptotic cells increased in MCF-7 cell lines transfected with SLC9A3-AS1- siRNA through the Annexin V-FITC kit and TUNEL kit
