11.SUMMARYREDUCING EFFICIENCY OF POLYENYLPHOSPHATIDYLCHOLINE ONRENAL DAMAGE IN OBSTRUCTIVE NEPHROPATHYPurpose: Obstructive nephropathy is the damage of the kidneys due to an abnormality ofthe urine flow in the urinary tract. Many congenital and acquired diseases of urinary tract causean obstructive nephropathy during childhood. Tubulointerstitial fibrosis is the characteristicfinding of the obstructive nephropathy and it has been demonstrated that oxidative stress have animportant role in the ethiopathogenesis of the obstructive nephropathy. Clinical and experimentalstudies have demonstrated that PPC have an antioxidant, antifibrotic and cytoprotective effects.In this experimental study, we aimed to investigate whether PPC treatment has any beneficialeffect on renal injury in unilateral partial ureteral obstruction (UUO) in rats.Material and method: Fourty Wistar-Albino rats were split into three groups (shamoperated controls, untreated and treated rats). Rats of the untreated and treated groups (n=15)underwent unilateral partial ureteral obstruction with the two-thirds of left ureter was embeddedrdin the psoas muscle. In the 3 , PPC was given orally at a dose of 100 mg/day for 30 days. At theend of 30-day experimental period, obstructed kidneys and blood samples were harvested. Toinvestigate the therapeutic efficacy of PPC treatment, oxidant and anti-oxidant enzyme levels(total nitrite and nitrate, glutathione, superoxide dismutase), lipid peroxidation(malondialdehyde), proinflammatory cytokines (interleukin-1α, interleukin-6, tumor necrosisfactor alpha), transforming growth factor beta-1, alpha-smooth muscle actin (α-SMA) andnuclear factor kappa beta expression, neutrophil infiltration, and tubulointerstitial damage in theobstructed kidneys were studied.53Results: PPC was found to be effective in decreasing the renal damage in obstructivenephropathy, and to decrease tubulointerstitial fibrosis. It was also found that PPC decreased theproinflammatory cytokines, growth factors and oxidant enzymes while increasing the antioxidantenzymes.Conclusion: We found that PPC decreases the renal damage after UUO, throughdecreasing the levels of TGF-β , NF-κβ and proinflammatory cytokines in addition to its strong1antioxidant, anti-inflammatory, antifibrotic and cytoprotective effects. We think that PPC cantake place in clinical use for the treatment of UPJ obstruction, PUV, neuropathic bladder andureteroceles, which are common pathologies of childhood period resulting in obstructivenephropathy.Key words: Partial unilateral ureteral obstruction, polyenyphosphatidycholine, totalnitrite and nitrate, neutrophil infiltration, proinflamatuar cytokines, transforming growth factor-beta1, alpha-smooth muscle actin, nuclear factor kappa beta
