Evaluation of DNA damage and oxidative stress in blood serum values of patients diagnosed with polycystic over syndrome

Abstract (EN)
Objective: It was aimed to evaluate oxidative stress and DNA damage in the blood serum of women with and without biochemical androgen elevation in women with polycystic ovary syndrome (PCOS). Materials and Methods: The study was conducted in Bezmialem Vakif University Hospital between April 2022 and November 2022 as a prospective cohort. Women between the ages of 16-40 diagnosed according to the Rotterdam criteria and healthy control patients were included. A total of 178 patients were included in the study and the patients were divided into 3 groups. The first group consists of women with PCOS without androgen elevation, the second group consists of women with PCOS with high androgen elevations, and the third group consists of healthy control patients. 2-5 of the menstrual cycles of all patients. blood tests were requested. Hormonal, metabolic and lipid parameters were measured from these samples, and also the presence of oxidative stress and DNA damage from blood serums was evaluated. At the end of the study, the effect of biochemical androgen elevation on oxidative stress and DNA damage in women with PCOS and its relationship with metabolic parameters were investigated. Results: A total of 178 patients were included in the study. According to the Rotterdam criteria, 112 PCOS patients and 66 control patients were recruited. The PCOS patient group was divided into two groups with and without androgen elevation. Among the groups, BMI was the highest in the hyperandrogenemic PCOS group. As a result of the study, oxidative stress damage, inflammatory marker and DNA damage values were found to be increased in the hyperandrogenemic PCOS group (p<0.05). In our study, no relationship was found between androgen elevation and metabolic parameters. Conclusion: In our study, we detected increased oxidative stress damage, chronic inflammation and DNA damage in patients with biochemical hyperandrogenemic PCOS. We found that women with nonadrogenic PCOS also had increased oxidative stress, chronic inflammation, and DNA damage when compared with the healthy group. However, we could not detect a significant relationship between androgen elevation and metabolic parameters. No significant difference was observed between PCOS and metabolic syndrome and lipid profile. Key words: PCOS, hyperandrogenemia, DNA damage, oxidative damage, chronic inflammation
