Abstract (EN)

In this study, a novel amperometric biosensor has been developed for the purpose of uric acid detection. While pyrole is being electropolimerized, paratoluensulfonate has been carried on the electrode surface together with the enzyme, meanwhile the enzyme has been immobilized via the entrapment method. As a result, Pt/polypyrrole-paratoluensulfonate-enzyme electrode has been prepared. Uric acid detection is based on the oxidation of hydrogen peroxide at +0.30 V produced by the enzymatic reaction on the enzyme electrode surface. The effects of pH, temperature and substrate concentration on uric acid biosensor response have been investigated. The linear working range has been found to be 5.10-7 - 5.10-5 M. KM and Vmax values of the immobilized enzyme have been calculated. The reusability and shelf life of the biosensor have been specified. The uric acid detection within blood has been achieved using the newly prepared electrode. The effects of the interference possibly occuring in biological environments on the biosensor response have been investigated.
