Investigation of the antioxidant and antidiabetic activities of prebiotic, postbiotic and paraprobiotic of potential probiotic levilactobacillus brevis KMP1 bacteria

Abstract (EN)

In this study, antioxidant activities of prebiotic L-EPS-KMP1, probiotic KMP1, postbiotic KMP1 and paraprobiotic KMP1 obtained from Levilactobacillus brevis KMP1 strain isolated from Balıkesir Kelle Mihaliç Cheese, one of the traditional Turkish cheeses, were determined by 7 different methods and IC50 values were calculated for antioxidant activity experiments. In addition, the ability to inhibit the α-amylase enzyme was investigated in order to determine the antidiabetic activity of prebiotic L-EPS-KMP1, probiotic KMP1, postbiotic KMP1 and paraprobiotic KMP1. The highest antioxidant activity in prebiotic L-EPS-KMP1 with ABTS+ scavenging activity method at 4 mg/mL concentration 66.88±4.4% and with superoxide anion scavenging activity test at 10 mg/mL concentration 56.21±3.45% has been determined. The lowest concentration of prebiotic L-EPS-KMP1 was 0.05 mg/mL, and the highest antidiabetic activity was 65.19±0.65%. The highest antidiabetic activity was found to be 92.12%±1.78% and 96.80±3.49% at 1:2 and 1:5 dilutions of postbiotic KMP1, respectively. Paraprobiotic KMP1 and its α-amylase inhibition activity in 1:2 and 1:5 dilutions were determined as 35.44±5.76%, 51.79±5.95% and 84.70±2.5%, respectively. With the methods applied in the study, it was determined that probiotic KMP1 strain, prebiotic L-EPS-KMP1, postbiotic KMP1 and paraprobiotic KMP1 showed antioxidant antioxidant properties, and prebiotic L-EPS-KMP1 had a protective effect against DNA damage. It is predicted by studies that prebiotic EPS, postbiotics, paraprobiotics and probiotics obtained from L. brevis KMP1 can be used as supplements in foods, can be used as a natural preservative in the field of food, and can be used as a natural antioxidant that makes an additional contribution to the microflora in the field of health. In addition, the results obtained from studies with postbiotics and paraprobiotics of prebiotic EPS obtained from L. brevis KMP1 show that it can be used as an alternative natural component to drugs in antidiabetic treatments.
