The investigation of relationship between protein oxidation and serum levels of nitric oxide, asymmetric dimethylarginine and homocysteine in pregnant women with preeclampsia

Preeclampsia may reflected a damaged NO metabolism table. Pathophysiologic mechanism that is characteristic of normal pregnancy and at least partially regulated with NO vasodilatation?s defficiency and decreased vascular reactivity insufficiency. It is thougt that one of the factors that play role in the preeclampsia pathogenesis is endothelial dysfunction. But it?s mechanism has not been exactly enlightened. Clinical and experimental studies is related the endothelial dysfunction with increased free radical production. Also, because of the inhibition of NOS activity with ADMA leads to a decrease in NO levels, it is expressed that endothelial dysfunction may develop as a result of this. One of the most important mechanisms in the reduction of nitric oxide function- as a result of homocysteine otooxidation- is the activation of oxidant system by production of reactive oxygen radicals and reduces NO bioavailability.We, in this study, in preeclampsia women, it is thought that play an important role in vascular homeostasis; NO level, evaluated as an endothelial dysfunction marker; ADMA and homocysteine levels and as an oxidative stres marker; protein carbonil levels measured.We did this study on 30 preeclampsia and 30 healty pregnant women, serum ADMA, SDMA and homocysteine levels were measured with HPLC method, serum protein carbonyl levels spectrophotometrically and NO levels were measured by Griess method with colorimetry.Our results showed us, in preeclampsia patient group according to control group serum ADMA, SDMA and homocysteine levels are significantly high (P<0.05). NO levels are significantly low (P<0.05). In consequence of statistical analyzes of the study serum NO with ADMA and between SDMA levels weak negative correlation, between ADMA and SDMA with PCO weak positive correlation were observed. Furthermore, between serum ADMA with SDMA and ADMA with homocysteine and SDMA with homocysteine levels strong positive correlation were determined.To do more large scale studies, in determination of the early diagnosis and prognosis of preeclampsia, we think that may be important when these parameters appear as new markers, both in prevention of probable complications and developing in new therapy approach.
