Correlation between microvessel density, expression of Ki-67 and p53 with clinicopathologic parameters and patient prognosis in prostate cancer

Abstract (EN)
SUMMARYWe investigated the prognostic value of microvessel density (MVD), Ki-67 stainingintensity and existence of p53 mutation together with clinicopathologic parameters in patientswho underwent radical retropubic prostatectomy (RRP) due to the localized prostate cancer.In our clinic, pathological specimens of 69 patients who have clinical and pathologicaldatas were evaluated from the patient groups who underwent RRP only, retrospectively. Thearea with the greatest density of each of CD31, Factor VIII and CD105 positive cell clusterswere chosen separately for counting MVD in X200 magnification. Greater than 5% of thetumor cells which demonstrated p53 nuclear reactivity were graded as positive. Thepercentage of nuclear Ki67 staining was grouped as negative, 1-10% and more than 10%.Mean follow-up of the patients was 37 (17-106) months. Disease progression wasdetermined in 17 (24.6%) patients during follow-up. Distribution of pathological stage inprogressive patients was more advanced than non-progressive counterpart (p=0,01).Transitional zone PSA density (TZPSAD) of non-progressive patients was statistically lowerthan progressive patients (p=0.01). Multivariate cox regression analysis revealed pathologicstage, Gleason score (GS) and TZPSAD as the independent predictor of progression freesurvival. Kaplan Meier analysis revealed higher pathologic stage and GS was significantlyshortened the time to progression (p=0.003,p<0.001,respectively).As a conclusion, MVD, percentage of Ki-67 staining and p53 protein expression donot add any predictive value in regard to progression after RRP compared with prognosticmarkers as pathologic stage and GS.
