Abstract (EN)

Title: Determination Of Pyroptosis Associated Genetic Variants And Protein Levels In Prostate Cancer Objective: Our study was conducted by looking at the protein levels of Gasdermin A, Gasdermin B, Gasdermin C and Gasdermin D, a total of four genetic variants of the pyroptosis-associated Gasdermin A (rs7212938), Gasdermin B (rs7216389), Gasdermin C (rs4527833) and Gasdermin D (rs2305492) genes. We aimed to present its effect on cancer pathology and new treatment options for the disease in question. Recently, pyroptosis has attracted attention due to its association with various types of cancer. It plays a common role in the emergence and development of various inflammatory diseases such as the progression of hepatitis to liver cancer and colonitis to colon cancer. However, a study showing the relationship between pyroptosis and prostate cancer has not been conducted yet. Methods: In our study, 100 patients who applied to ESOGU Faculty of Medicine, Department of Urology were diagnosed with prostate cancer, and 120 healthy control individuals who applied with various complaints and no malignancy were detected were included. SNP-Genotyping for variant analysis of Gasdermin family genes and ELISA method for protein levels were used and the results were evaluated statistically. Results and Conclusion: As a result of our study, when the pyroptosis-associated GSDMD (rs2305492) genetic variant was examined in terms of allele frequencies, a significant difference was determined between prostate cancer patients and control individuals in terms of A allele (P=0.007). Again, in terms of genotype variables, a significant difference was determined between prostate cancer patients and control individuals for the GSDMD genetic variant (P=0.016). In terms of protein level, a significant difference was determined between GSDMC, prostate cancer and control groups (P=0.013). Our findings may contribute to the creation of new strategies for the diagnosis and treatment of prostate cancer and further studies.
