Abstract (EN)

Serum proteins are used for the determination of biomarkers which provide the diagnosis of diseases. Because albumin and immunoglobulin G (IgG) concentration in blood serum is very high, serum proteins prevent the determination of biomarkers which are at low concentrations. Therefore; the proteins depletion of albumin and IgG which comprises approximately 80 % of the serum, becomes important. In this thesis, primarily, the depletion of albumin and IgG have been carried out with nanoparticle systems. For this purpose; three different nanoparticles with the binding of Protein A (Pro A), Cibacron Blue F3GA (CB) and both Pro A and CB to Fe2O3 nanoparticles have been prepared. These nanoparticles have characterized and depletion of albumin and IgG have been carried out from aqueous solutions and serum. It has observed that the solid-phase systems are able to deplete of albumin and IgG effectively and in a short time. In the second stage of the study, the solid phase systems have prepared for the determination of cancer antigen (CA 125) and lysophosphatidic acid (LPA) which are used as biomarkers for ovarian cancer. The nanosensors which have prepared via imprinting of phosphoserine and LPA onto carbon nanotube (CNT) and Fe2O3 nanoparticle have synthesized, characterized and detection limits of CA 125 and LPA have determined.
