Abstract (EN)
Protocatechuic acid, found in many medicinal plants, is a phenolic humectant known for its anti-inflammatory, analgesic, antioxidant and neuroprotective activities. Within the scope of the thesis, the aim is to elucidate the effect package that shows the neuroprotective breakdown of protocatechuic acid into the anxiety and reproducible examples of behaviors in which we expect cuts in wages as a result of LPS-induced results. Tail tuning test (depression), text-darkness test (anxiety) and open field test (locomotor activity) were used to score LPS-induced mood scores in mice. The changes in IL-1β, IL-6 and TNF-α levels in mice administered LPS are different from the effects of protocatechuic acids on these cytokine levels. In order to elucidate the possible consequences of LPS-induced systemic treatment of protocatechuic acid via the TLR-4/NFκB pathway, the expressions of TLR4 and NFκB at the mRNA level are determined. In this study, the presence of protocatechuic acids and their possible beneficial effects on anxiety disorders were evaluated for the first time in an inflammatory disease model, focusing on neuroimmune therapy. In addition, with our study, the role of the TLR-4/NFκB pathway in the possible neuroprotective effects of protocatechuic acid was examined in vivo for the first time. Keywords: Protocatechuic acid, LPS, Inflammation, TLR4, Anxiety.
