Investigation of the relationship between chromosome aberrations and micronuclei formation after irradiation

DNA molecule damage is caused by hereditary or environmental factors. These damage start the carcinogenesis process. The usefulness of chromosomal aberrations as DNA cytogenetic markers in the detection of DNA damage brings the use of techniques that measure these occurences to the diagnosis of cancer. In this thesis, micronucleus and non-permanent chromosomal aberration techniques which measure chromosomal aberrations are compared and investigated in terms of similarities and differences in their response to radiation, a carcinogenic factor. For this purpose, in vitro cells were irradiated with radiation. The relationship between the frequencies of micronucleus and non-permanent chromosome aberrations was compared in terms of dose-response curves. Dose-response curves were determined with TK6 lymphoblast cells, a human cell line. One of the cells growing in different cell culture flasks was separated as control and the others received 1 Gy, 2 Gy, 3 Gy, 4 Gy radiation dose in vitro. Dicentric and free asentric frequencies obtained after doses were determined by chromosome aberration technique. Micronucleus (MN) frequencies were determined by micronucleus technique. Statistically optimal dose-response curves were generated. The relationship between the curves is examined. In this thesis, graphs are plotted as second order polynomials using non-linear feature of regression analysis. The data obtained showed that the increase in the radiation dose increased the MN/BN ratio and increased radiation specific dicentric and asentric fragments. Increases in radiation dose showed a significant differences (p<0,05) between MN and chromosome aberrations.
