The effect of resveratrol on endometriotic implants in the rat model

Endometriosis is defined as the presence of endometrial tissue, consisting of both glandular epithelium and stroma, outside the uterine cavity. This condition is predominantly found in women of reproductive age with chronic cyclic or noncyclic pelvic pain, dysmenoure, dyspareunia, and infertility. Endometriosis is an estrogen-dependent condition. Therefore, medical therapy is often aimed at lowering estrogen level. However, side effects occuring due to hipoostrojenizm restarins its long term use.Along with various mechanisms held responsible of endometriozis formation, it is also shown that anjiogenezis plays an important role in formation of endometriozis and inflamatuar reaction in its continuation. Goal of our study is to research the effects of resveratrol which shows anti-inflamatuar and anjiogenezis inhibiting effect in rat endometriozis model.Twenty-four adult female rats were used and all rats underwent three operations laparotomies. The ectopic endometrium was induced by the first surgery. the surface areas of the implants were measured and peritoneal lavage was performed in the second laparatomy. the rats were randomly divided into three groups after the second surgery. Group I (reveratrol group) were given 60mg/kg/day (peroral,21 day).Group II were given a single dose of 1 mg/kg s.c. leuprolide acetate (GnRH agonist group). Group III were given no medication and served as controls. After the end of the medical treatments the third laparotomy was performed. the surface areas of the implants were measured, peritoneal lavage was performed and the implants were then excised and fixed in10% formalin for histopathological examination in the third laparatomy.At the end of the treatment, the mean areas of implants were statistically significant smaller, VEGF and MCP-1 levels in peritoneal fluid were statistically significant lower in Groups I and II when compared with the control group. The mean score of the histopathological examination of the implants and VEGF and MCP-1 levels in blood at the end of the treatment was statistically significant lower in Groups I and II, when compared with (Group III) the control groupIt is shown in our study that in rat endometriozis model decreasing VEGF and MCP-1 in peritoneal liquid resvatrol provides shrinkage in endometriotic focals by creating anti inflammatory effect and affecting anjiogenezis in a negative way.We think that in rat endometriozis model our study, which researches the effects of resveratrol on endometriotic focals, can lead other studies to be done on resveratrol and endometriozis and can be a new hope in endiometriozis treatment.Key words: resveratrol, angiogenesis, endometriosis, vascular endothelial growth factor, monosit kemotactic peptit
