The compared effects of Relcovaptan and GNRH antagonist in an ovarian hyperstimulation rat model

More couples are being affected by infertirity due to the rise in the age of conceiving and diseases that cause infertility such as pelvic inflamatory disease..Artificial reproductive technologies are used widely and ovarian hyperstimulation syndrome is encountered in approximately 3-6% of cycles that ovarian hyperstimulation is applied. Because of vascular hyperpermeability; this syndrom which has a clinical presentation that is characterized with free fluid and pleural effusion, the primary suspicious agent is vascular endothelial growth factor (VEGF). There is an opinion that there are many other factors that act in pathogenesis of ovarian hyperstimulation syndrom.One of that factors is arginin vasopressin that stimulate VEGF potently and plays an important role on vascular permeability.The aim of our study was; block the vasopressin V1a receptors via inhibiting VEGF by Relcovaptan®(SR49059) and prevent OHSS generation. We used female immature Wistar rats as the OHSS experimental model. In total 30 rats were incorporated in the study and they were split in to 5 groups. 6 were73selected as the control group and only saline infusion was applied during the study. PMSG used in the remaining 4 groups for ovarian hyperstimulation and hCG was applied on the 5th day of study. On the day of hCG and the following day GnRH antagonist was applied twice each day and Relcovaptan was applied with different doses (0.3 mg, 0.15 mg) to the other two groups. No treatment was applied to 4th group that was thr OHSS group.Groups were compared with their body weight, total ovarian weight, VEGF amounts in peritoneal fluid, corpus luteum, antral and atretic follicule counts.According to the results of the study; in each treatment group mainly peritoneal VEGF levels, body weights and the total ovarian weights were significantly decreased statistically when compared with the OHSS group.Key words: Relcovaptan, OHSS, VEGF, Rat, Vasopressin inhibitors
