The protective effect of cb2 agonist against ovarian ischemia/reperfusion injury in rats

Abstract (EN)

Aim: Ovarian ischemia/reperfusion injury is one of the cases of acute abdomen during reproductive period in women and one of the most common causes is ovarian torsion. If left untreated, necrosis may develop due to decreased blood flow to the ovaries and the organ may lose function. The aim of thesis is to evaluate the protective effect of cannabinoids in ovarian torsion injury and to investigate the role of CB2 receptor in experimental ischemia/reperfusion injury in rat ovaries. Material and method: In this study, 28 female wistar rats were used. The rats were divided into 5 groups as sham control group, ischemia / reperfusion group, CB2 agonist+ischemia reperfusion group, CB2 agonist+antagonist+ischemia/reperfusion group and ischemia/reperfusion+dimethylsulfoxide group. Ovarian tissue removed. MDA, GSH, TNF-α and IL-1β levels were measured in tissue parts. These levels were compared to sham control group. Results: I/R group (3066,67±381,66, pg/ml) compared to sham control (823,33±206,56, pg/ml) serum TNF-α levels increased. Agonist reduced ovarian I/R damage as to serum TNF-α (1434,67±400,91, pg/ml). Differences were found when I/R group was compared to sham group (P<0,001) according to IL-1β level. When agonist group (8,92±1,33, pg/g) and antagonist group (13,01±2,45, pg/g) were compared, the result was statistically significant (P=0,046). When the MDA level was compared to sham group (77,66±12,28, nmol/g) and I/R group (109,06±11,04, nmol/g), it was found that I/R caused an increase in MDA level and CB2 agonist (86,50±16,32, nmol/g) was found to cause a decrease in MDA level. The effect of agonist was not affected by the antagonist (99,11±5,07, nmol/g, P=0,39). When I/R group (24,51±6,06, mg/100 ml, 0,56±0,25, µmol/g) and sham control group (38,06±5,08, mg/100 ml, 1,33±0,07, µmol/g) were compared in terms of GSH levels, GSH levels were found to be decreased in serum and tissue. Conclusion: CB2 receptor may support treatment of ischemia reperfusion injury in ovary.
