Effect of toll-like receptor antagonist eritoran in retinochoroidal inflammatory damage at rat model of endotoxin induced sepsis

Toll-like receptors play a key role at sepsis which is characterized by uncontrolled systemic inflammatory response. These receptor family members recognize different microbial structures whereas TLR4 recognize gram negative bacteria lipopolysaccharide. TLR4 molecule stimulated by LPS trigger some reactions and finally cause proinflammatory mediator release via nuclear factor kappa stimulation. TLR4 molecule especially found on immun system cells but also found on some ocular cellular structures. TLR4 antagonist Eritoran is a synthetic lipid A analogue and prevents the engagement of LPS and TLR4. The aim of this study was to evaluate the biochemical and histopathologic changes at retinochoroidal tissue in endotoxin induced sepsis rats and the improvement in retinochoroidal inflammatory damage with eritoran, TLR4 antagonist.This study was held in Gazi University Test Animal Laboratory. Rats are studies in five groups 10 rats found in each. First group was sham group for evaluation of normal value of parameters. Control and study groups were assessed parameters in 12th and 24th hour. While control group rats were delivered saline solution, study groups took eritoran through tail vein. According to the different timing of groups blood samples were collected and enucleation were done at 12. and 24. hours. TNF-?, MDA, NF?B levels were evaluated at serum and retinochoroidal tissue and also histopathological assessment were made.At 12th hour of study an improvement was seen in all parameters but this change was not significant istatistically. At 24. hour although sepsis parameters were worsened all parameters improved significantly. Serum TNF-? and NF?B levels were not different than normal sham rats at 24th hour.Eritoran, TLR4 antagonist, provide improvement in inflammatory damage both in serum and retinochoroidal tissue parameters evaluated by TNF-?, MDA and NF?B levels especially at 24th hour of the study. Future investigation needed in transport of eritoran from ocular barriers, toxicity and dosage.
