Abstract (EN)
Microbiome is one of the most complex components of the human body. The intestinal flora called the second brain contains the largest number and variety of microrganism. Drugs that enable the treatment of diseases and diseases have an impact on microbiology and metabolism. The aim of this thesis is to reveal the effects of two different doses of reboxetine, one of the selective noradrenaline reuptake inhibitors, and the effect of healthy-diabetic control groups on the gut flora with bioinformatics analysis. After the administration of 8 mg/kg and 16 mg/kg reboxetine to diabetic and healthy rats, DNA was isolated from stool samples. V3-V4 primers specific to the 16S rRNA region were used for the application procedure and sequencing was performed with Illumina Miseq. After the preprocesses, alpha diversity beta diversity and relative taxa distributions were determined by using QIIME 1.90 bioinformatics analysis program. There was no difference in alpha metrics. Beta diversity was found statistically significant in all experimental groups, diabetic-healthy control groups, and in drug doses administered to diabetics (p<0,005). After the LefSe analyzes, Lactobacillus, Turicibacter, Streptococcus significantly increased (p<0,05). The analyze showed that Parabacteroides, Bacteroides, Flexispira and Desulfovibrio increase significantly (p<0,05) in the group treated with reboxetine. According to the results, it was shown that reboxetine may cause dysbiosis similar to diabetic cases in microbiota. Keywords: Reboxetin, Gut flora, Bioinformatics microbiata analysis.
