Investigation of Biochemical Response in Rainbowtrout (Oncorhynchus mykiss) Exposedto Remazol Brilliant Blue R Textile Dye The dyes used in textile industry production are toxic substances and most of them have polymer structures. These dyes pollute the aquatic environment via various stages such as printing and dyeing, and may cause molecular, cellular, biochemical and physiological disorders. In this study, it was aimed to investigate the response of Rainbow Trout exposed to Remazol Brilliant Blue R, which is widely used in textile industry in varying concentrations. With this purpose, 56 Oncorhynchus mykiss (Rainbow Trout) individuals were obtained from local farms. The Rainbow Trout (Oncorhynchus mykiss) were applied a dyeing substance in an experimental setting with a control group and three experiment groups. The Glutathione S-transferase (GST), Cytochrome P-450 (CYP1A1) and Lactate Dehydrogenase (LDH) enzyme activities of these groups were measured. These measurements were read in a microplate reader using commercial kits with ELISA method. In comparison with the control group, the GST enzyme activity increased in the groups B (1 mgl-1) and C (2 mgl-1) in the 24th hour, and the difference between the groups was significant (p<0.05). Increases were observed in the groups A (0,5 mg l-1) and B (1 mg l-1) in the 48th hour (p<0.05). In comparison with T0, the CYP1A1 enzyme activity in the group B (1 mg l-1) increased by the 24th hour, and decreased by the 48th hour (p>0.05). In comparisons of the groups with different dosages and the control group in the 24th hour, CYP1A1 enzyme activity increased in the groups A (0,5 mg l-1) and B (1 mg l-1) (p<0.05), and decreased in the group C (2 mg l-1) (p<0.05).In comparisons of the groups with different dosages and the control group in the 24th hour, LDH enzyme activity showed a significant increase in the groups A (0,5 mg l-1), B (1 mgl-1) and C (2 mg l-1) (p<0.05). When compared to the control group in the 48th hour, the groups A (0,5 mgl-1) and B (1 mg l-1) showed a significant increase (p>0.05). According to the results of this study, it was shown that CST, CYP1A1 and LDH enzymes may be useful as biomarkers in determining the toxic effects in Oncorhynchus mykiss (Rainbow Trout) exposed to the RBBR textile dye.
