Abstract (EN)

Role of Resveratrol On The Effect of Resistin On Rabbit Corpus Cavernosum Türkan GÜNGÖR. Dokuz Eylul University Institute of Health Sciences, Department of Pharmacology. 35340 Balcova-Izmir Objective The most important reason of erectile dysfunction (ED) is endothelial dysfunction (ED). Resveratrol improves the ED in the penile tissue due to hypercholesterolaemia. It has not been investigated whether the resistance of endothelial function defect in the cardiovascular system (CVS) has a role in ED. For this purpose, it was investigated that the physiopathological effect of resistin on erectile tissue was directly determined in vitro conditions and this effect would be reversed with resveratrol. Method In our project, 19 rabbits weighting 2000-3000 g were used and in vitro organ bath method was used to evaluate endothelium-dependent relaxation responses with acetylcholine (Ach), endothelin-1 mediated contractile responses and endothelium-free relaxation responses with sodium nitropuriside (SNP) the corpus cavernosum tissues incubated with krebs, resistin and resistin +resveratrol for 24 hours. In these groups, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, which is known to increase in oxidative stress was measured. Results After 24 hours of incubation and in order to demonstrate the viability of the tissues at the end of the experimental protocol, 120 mM potassium chloride (KCl) was applied to the tissues and no significant difference was observed between KCl-induced contractile responses. There was no significant difference between groups in terms of relaxation responses generated by Ach and SNP after phenylephrine (Phe) contraction and endothelin-1 mediated contraction responses for Krebs group, resistin group and resistin + resveratrol groups. Resveratrol administered in the presence of resistin significantly reduced NADPH oxidase activity compared to the group treated with resistin alone. Conclusion Resistin may have the potential to alter NADPH oxidase activity, and resveratrol may be an alternative treatment target for ED.
