Assessment of reproductive toxicity of risperidone in male rats

Abstract (EN)

It is known that risperidone (RIS), a commonly used drug in the treatment of schizophrenia and mania, causes some adverse effects in the male reproductive system, but there is no comprehensive study evaluating possible reproductive toxicity of RIS within its mechanisms. In this study, male rats were administered orally for 1.25, 2.5 and 3 mg / kg RIS for 28 days and the sperm concentration, motility, morphology, DNA damage and the histological changes in testicular tissue were evaluated to determine the reproductive toxicity of RIS. In addition, follicle stimulating hormone (FSH), luteinizing hormone (LH) and serum levels of testosterone which plays the main role in the hormonal regulation of reproductive functions of and the underlying mechanisms of oxidative stress among the underlying mechanisms and oxidative stress glutathione (GSH), catalase (KAT), superoxide dismutase (SOD) and malondialdehyde (MDA) levels were determined in testicular tissue. According to the results, it was observed that normal sperm morphology was decreased in all dose groups compared to the control group and histopathological changes occurred in testis tissue dose-dependently. Serum LH levels were not altered in the RIS groups, and FSH and testosterone levels decreased in the highest dose group compared to the control group. In addition, the highest dose of RIS group in the testicular tissue GSH levels were seen to decrease, MDA levels wereincreased which were evaluated as indicators of oxidative stress. As a result, RIS caused reproductive toxic effects in male rats by inducing oxidative stress and disrupting hormonal regulation. Keywords: Risperidone, Sperm, FSH, Testosterone, Oksidatif stress.
