Abstract (EN)

Rutin has been known that is an flavonoid which has analgesic and anti-inflammatory activity and is found in many medicinal plants. In this thesis, it was aimed to evaluate the analgesic efficacy of routine with hot plate and tail immersion tests in mice and to reveal pain pathways involved in this effect. Rutin doses of 25, 50, 100 and 200 mg/kg (i.p.) were administered to mice to evaluate the analgesic effect, and 300 mg/kg (i.p.) dipiron and 30 mg/kg (i.p.) tramadol were used as positive controls. In order to examine the pathways involved in the analgesic effect of rutin; Experiments were repeated using the alpha-1 adrenergic receptor antagonist prazosin and the alpha-2 adrenergic receptor antagonist yohimbine, serotonergic receptor antagonists ketanserin (5-HT2), ondansetron (5-HT3) and WAY100635 (5-HT1) and, the opioid receptor antagonist naloxone. In addition, the analgesic effect of rutin and tramadol combination was evaluated in this thesis study. It was determined that there was a significant analgesic effect in all doses of rutin. When we evaluated the analgesic effect of 100mg / kg routine in the presence of antagonists, it was determined that serotonin and opioid receptors mediate this effect. The analgesic effect of the combined use of routine and tramadol was found to be lower than the effects of both drugs when administered alone. As a result, we can state that it is a flavonoid with potential to be evaluated in new drug development studies due to its routine analgesic effect.
