Özet (EN)

Ammonium Pyrrolidine Dithiocarbamate (APDTC) is the strongest inhibitor of NF-κB. In addition to this property, antitumoral, antioxidant, anticancerogenic, antiviral, metal chelator and inhibitory effect on smooth muscle cells apoptosis were determined. In our study, our aim was to investigate the mechanism of action of APDTC on ileum longitudinal smooth muscle. Adrenergic receptors, cholinergic receptors, L-type calcium channels and potassium channels are blocked in different groups to determine how effective these channels and receptors are on this mechanism. In this study, contraction-relaxation responses to APDTC were investigated after administration of atropine, phentolamine, propranolol, nifedipine, tetraethylammonium (TEA) and atropine+ phentolamine+ propranolol and control in acetylcholine precontracted ileal tissue. APDTC forms relaxation responses on ileum. Atropine did not change the relexantion response of APDTC. Phentolamine did not change the relaxantion responses of APDTC. However, it significantly inhibited the contraction induced by acetylcholine. Proporanolol did not change the relaxantion effect of APDTC. APDTC has a relaxing effect on propranolol in response to acetylcholine-induced contraction response. Nifedipine has not changed the relaxantion response of the APDTC. TEA has not changed the relaxantion effect of APDTC. TEA increased much more the response to acetylcholine- induced contraction. To determine the NANC status, adrenergic and cholinergic receptor blockade done at the same time furter enhanced the relaxation effect of APDTC. It has been determined that APDTC is not effective on NANC system. As a result, it is thought that APDTC, which we applied on the rat ileum smooth muscle, acted by different receptors and pathways besides the receptor antagonists and channel blockers that we use in our study.
