The investigation of action mechanism of ammonium pyrrolidine dithiocarbamate on rat bladder smooth muscle contraction-relaxation responses

The specific inhibitor of NF-κB, ammonium pyrrolidine dithiocarbamate (PDTC), anti-viral, anti-apoptotic, anti-inflammatory and antioxidant effects is a proven thiol compound in many studies. There is not enough literature on the ways in which APDTC has produced these effects. In our study, it was aimed to investigate the mechanism of action of APDTC in in vitro rat bladder smooth muscle. In our study, 70 Wistar albino male rats were used. The bladders of the rats which were euthanized by cervical dislocation under the anesthesia were opened and their bladders were isolated. After the removal of the connective tissues around the bladders, they were placed in the organ bath with Krebs solution. KCl, ACh, APDTC, Atropine, Fentolamine, Propranolol, Nifedipine, TEA were applied to the bath with an appropriate protocol. The data were evaluated with Kruskal Wallis and Mann-Whitney U tests. ACh induced bladder induced APDTC relaxation response in smooth muscle. Cholinergic receptor blocker Atropine, β-adrenergic receptor blocker Propranolol, L-type calcium channel blocker has not changed the relaxation response of mix APDTC consisting of nifedipine and atropine + phentolamine + propranolol. In the presence of potassium channel blocker TEA, APDTC produced contraction response, but this was not a significant response. α-adrenergic receptor blocker In the presence of phentolamine, APDTC produced significant contractile response. As a result, it is thought that APDTC is effective in α-adrenergic receptors and NANC system in rat bladder smooth muscle.
