The effect of dexmedetomidine against renal ischemia reperfusion induced oxidative damage in rats

Abstract (EN)
Aim: We studied the protective effect two different doses of dexmedetomidine, an alpha-2 adrenergic receptor agonist, against oxidative damage during renal ischemia-reperfusion in rats. At the same time we examined their effects on levels of BUN, creatinin (Cre) and TNF-?.Materials and Methods: Male Sprague-Dawley rats were divided into 4 groups each including 10 animals: Group 1 (control), group 2 (ischemia-reperfusion), group 3 (dexmedetomidine 100 µg / kg) and group 4 (dexmedetomidine 10 µg / kg). After removing right kidney of rats, left kidney was performed ischemia during 40 minutes and reperfusion in following 3 hours. Biochemical analysis was performed on blood and left kidney tissue obtained at the end of the experiment.Results: In group 2, compared to the control group levels of BUN, Cre and kidney tissue malondialdehyde (MDA) increased significantly (p <0.05), superoxide dismutase (SOD), catalase (CAT) enzyme activity and glutathione levels decreased significantly (p <0.05). In group 3, compared to the group 2, levels of BUN, Cre decreased significantly (p <0.05), SOD activity was increased significantly (p <0.05). There was no significant difference between group 3 and group 4 in the all measures and Changes in the group 4, compared to the group 2 is found to be similar in the all measures. In the group 4, compared to the group 2, TNF-? levels were decreased significantly (p <0.05)Conclusion: Dexmedetomidine shows the antioxidant effects against renal ischemia reperfusion injury clearly at 100 µg/kg dose and 10 µg/kg.Key words: Rat, kidney, ischemia-reperfusion, oxidative stress, dexmedetomidine.
