Hasar grupları arasındaki doku SOD1, GPx, CAT, MDA, TAS, TOS, OSI değerleri karşılaştırıldığında farklılık saptanmamıştır (p > 0.05). Sinir lifi çapında Grup 1 ile 2 ve Grup 1 ile 3 arasında (p< 0.001); schwann hücre apoptozis skorunda Grup 1 ile 2 ve Grup 1 ile 3 arasında; aksonal dejenerasyonda Grup 1 ile 2 ve Grup 1 ile 3 arasında; myelin dejenerasyonda ise Grup 1 ile 2 ve Grup 1 ile 3 arasında istatistiksel olarak anlamlı farklılık saptanmıştır (p < 0.05). Sonuç Edaravonenin hayvan modelinde oluşturulan travmatik fasiyal sinir paralizilerinin tedavisinde etkili olduğu ve sinir fonksiyonlarındaki iyileşmenin histopatolojik sonuçlar ile de uyumlu olduğu gösterilmiştir. Anahtar Kelimeler Edaravone; Fasiyal paralizi; Ezilme yaralanması; Kornea refleksi; Apopitozis; Antioksidan etki SUMMARY The Evaluation of the Effect of Edaravone on Recovery of the Facial Nerve After Crush Injury in Rats Introduction Facial nerve paralysis is a significant disorder that leads to functional, cosmetic and psychological problems in individuals. Today, there is a need for new alternative therapies that increase nerve regeneration in addition to current surgical and medical treatment methods. In our experimental study, we aim to evaluate the effects of edaravone on healing in facial nerve crush injuries in an animal model. Materials and Methods The study was initiated after the consent of the ethics committee was obtained. The experimental study involved 30 Sprague-Dawley adult female rats weighing between from 250-300 gr rats were randomly divided into 3 groups of 10 rats. Immediately before branching of the facial nerve trunk, facial paralysis was created by crushing with mosquito clamp for approximately 1 minute. Afterwards i.p. SF, Prednol and Edaravone were administered. Functional evaluation was performed using whisker movements and corneal reflex at 1, 2, 3, 4 weeks. The extracted truncus was examined by histolopatologically. SOD1, GPX, CAT, MDA, TAS, TOS, OSI values were measured in the tissue samples of facial nerve branches and the serum. P <0.05 was accepted as statistically significant. Results Corneal reflex was evaluated and significant difference was found between all groups (Group 1, Group 2, Group 3) in the fourth week (p <0.005). There was no significant difference between Group 2 and Group 3 in the fourth week, but there was a significant difference between Group 1 and Group 3 (p = 0.003). In the fourth week, whisker movements were detected to be significantly different among all groups (Group 1, Group 2, Group 3) (p <0.005). There was no difference between Group 2 and Group 3 in the fourth week, but there was a difference between Group 1 and Group 3 (p = 0.029). A statistically significant difference was found between Group 2 and Group 3 when TAS and OSI levels were compared between serum groups. There was no difference in tissue SOD1, GPx, CAT, MDA, TAS, TOS, OSI values between the injured groups (p> 0.05). The histopathological evaluation was as follows: nerve fiber diameter; significant difference between groups 1 and 2 and between groups 1 and 3 (p < 0.001); schwann cell apoptosis score: significant difference between Group 1 and 2 and Group 1 and 3; axonal degeneration: significant difference between groups 1 and 2 and between groups 1 and 3; myelin degeneration: significant difference between groups 1 and 2 and between groups 1 and 3 (p < 0.005). Conclusions It was demonstrated that edaravone is effective in the treatment of traumatic facial paralysis in animal model and improvement in neural function is correlated with histopathological results.
