The effect of aca induced okadaic acid neurodegeneration in rats

Abstract (EN)

Aim: Recent studies ıt has been shown that the TRPM2 and PLA2 inhibitors may have a protective effect on neurons. This study was aimed to investigate the protective effect of TRPM2 and PLA2 inhibitor N-(p-amylcinnamoyl)anthranilic acid (ACA) in a neurodegenerative model induced by Okadaic acid (OKA). Material and Method: Male Sprague-Dawley rats were randomly divided into five group (n=10); 1) control, 2) sham: rats were injected icv with aCSF and treated vehicle for 13 day, 3) ACA: rats were treated with ACA ip 25 mg/kg/day for 13 days, 4) OKA: OKA was dissolved in aCSF and injected icv (200 ng) in a volume of 10 μl bilaterally and 5) OKA+ACA: rats injected icv with OKA (200 ng) and treated with ACA ip 25 mg/kg/day for 13 days. After 13 days of surgical operations and injections, Morris water maze test was performed. The histopathological finding, SOD, GSH-PX enzyme activation and MDA and GSH, TNF-α, IL-1β levels in tissues the hippocampus and cerebral cortex were measured. Results: The latency to platform and distance moved of OKA group rats were significantly increased in comparison to control, sham and ACA, OKA+ACA groups. In while the MDA, TNF-α levels of tissues were significantly increased in OKA group; SOD, GSH-PX enzyme activation and GSH levels were decreased compared to all of other groups (P< 0.05). OKA+ACA gruop comparison to OKA group, increased SOD and GSH-PX enzyme activation and GSH levels, and conversely decreased the levels of MDA, TNF-α. When OKA+ACA rats compared to OKA rats, ıt was found that numbers of degenerated neurons and caspase 3 positive cells of cortex and hippocampus regions were significantly reduced. Conclusion: ACA administration prevented to oxidative stres and neuroinflammation of OKA induced neurodegeneration. ACA ameliorated to cognitive decline and neurodegeneration.
