Obtaining, partial purification and characterisation of cellulase enzyme from bacteria isolated from Ağri Diyadin thermal hot springs

Abstract (EN)

In this study, the characterisation of cellulase enzyme of Bacillus licheniformis bacteria obtained from a thermal hot water source in Diyadin District of Ağrı Province, the effects of various compounds on the enzyme, the amount of exopolysaccharide produced by this bacterium and the antimicrobial effect of exopolysaccharide were investigated. The optimum operating temperature and pH of cellulase enzyme were determined as 60 oC and 9.0, respectively. According to these results, Vmax value was 2.33 EU/ml and Km value was 0.0348 mM for cellulase enzyme with carboxymethyl cellulose (CMC) substrate. When the effects of EDTA, KCl, MgSO4, PIO3 and SDS compounds on cellulase enzyme were analysed, these compounds inhibited the enzyme. IC50 and Ki values of these compounds were calculated by drawing Lineweaver-Burk graphs and inhibition types were determined. As a result of the studies, it was determined that Bacillis licheniformis bacteria grows at pH 6.0 and produces the most exopolysaccharide at this pH and 40 oC, and the exopolysaccharide produced has antimicrobial effect.
