Determination and relationship of some molecular marker polymorphisms affecting production traits in cattle breed

Abstract (EN)
In this study we aimed to determined molecular marker polymorphism in the ELF5 (E74-Like Factorr 5), BMP2 (Bone Morphogenetic Protein2) and FGF21 (Fibroblast Growth Factor 21) which effected the performance traits of cattle. We are used as material the Brown Swiss breed DNA samples. PCR analysis were made for the three different genes and then DNA samples were clustered by High Resolution Melting Analysis (HRM) method and sequence analysis were applied to some of the samples in each different cluster and were defined its polymorphic regions. In the result of DNA sequence analysis for ELF5, FGF21 ve BMP2 gene regions: for the 2. intron of ELF5 gene; g.65825371A deletion, g.65825359 C/T and g.65825499 G/A transistions, for the 5. exon region of ELF5 gene; G.65825909 T/C, g.65826062 G/A, g.65826123 A/C, g.65826134 C/T; and g. 65826138 T/C polymorphic region, for the 1. exon region of the FGF21 gene; G.55834828 C/G polymorphic region, for the 3. exon region of FGF21 gene; g.55836088 G/T and g.55836007 C/A polymorphisms, for the 2. exon region of BMP2 gene; g.9551963. T/C polymorphism and for the 3. exon region of BMP2 gene; g.49551337 G/A, g.49551405 G/T, g.49551428 G/C, g.49551433 A/C and g.49551449. T/G polymorphic regions were defined. There was non-significant as statistically the relationship between the polymorphic regions determined for the ELF5, BMP2 and FGF21 gene and lactation milk yields, 305-days milk yields, daily milk yields, lactation times, peak milk yields, peak day yields and average peak milk yields of Brown Swiss breed (P>0.05). However, it is suggested to investigate the marker ability of polymorphic regions determined for future studies in the animal breeding. Keywords: ELF5, BMP2 and FGF21, HRMA, polymorphism, milk yield
