Abstract (EN)
For many years, lipidic, polymeric or inorganic nanoparticles have been extensively investigated for the transport of various anticancer, antifungal, analgesic, drugs and important biological molecules within the body. However, a safe delivery system that is biocompatible and biodegradable, capable of remaining in the circulatory system for a long time without instability and toxicity, evading the immune system, remaining at the target site for a long time, has still not been found. Exosomes, which have been released from cells in recent years and act as natural carriers, can prevent the disadvantages of nanoparticle systems in these respects. In this thesis study, plant-derived exosomes obtained from lemon were used as an siRNA delivery system that could be effective in lung cancer cells. Within the scope of the study, characterization analyzes (NTA, DLS, zeta potential, TEM), protein content determination, immunophenotyping, transfection, siRNA loading and cell viability effects of siRNA-loaded exosomes on HUVEC, A549 and MCF-7 cells on exosomes obtained from lemon by three methods were evaluated as MTT and apoptotic effect level. As a result, with this thesis study, BCL-2 siRNA was loaded into lemon-derived exosomes for the first time and its effect was evaluated. As a result of the analyses, it is thought that it will pave the way for the development of plant-derived exosomes that can be used in cancer treatment as a candidate genetic material carrier system.
